Blocking is good, and necessary, as Grant mentioned.
Are you seeing long term blocking? Is there a specific issue, or did you just see blocks held in sp_who2?

The advice is
- domain user
- everyone group ONLY
- use SQL Server Configuration Manager to assign the account as the service account(s)
- add specific file share rights to access folders...

Log reader tools can help, but do you know what the large transaction was? If so, you can write the "reversing" transaction yourself.
