I personally wouldn't recommend either. The @variable IS NULL OR Column = @Variable produces poor execution plans (unless option recompile is specified and the SQL version is > 2008 SP2)...

Depends whether the query is resolved with an index scan or lots of index seeks. Check the execution plan.

If it is part of a large range scan from disk, maybe.

No, that would probably not be a particularly good thing to do. I'd recommend create a job that updates stats on that table with fullscan and schedule it every night.
