The only other option you have on a database level is the trustworthy option. If trustworthy=OFF then CLR assemblies marked unsafe or external access will not run.

Execute is a database level permission, so you'll have to create database level role for it.
Additionally, you cannot create server-level roles in SQL 2008.

Yup. Mostly. There maybe things that won't work on compat mode 90 that will on 100 because of syntax rules, but it won't be a case where all 2008 features...

It's probably worth mentioning that locking hints are not the way to solve blocking problems in a database.
