I went through a similar transition a couple years ago - moving an Access application to ASP (pre .NET) with SQL server as the backend. Luckily, the app was for an intranet - so I didn't have the added security concerns that you're likely to have. Opening a database to updates via the internet comes with a whole slew of security concerns.
For a starting point, I can point you to the resource that I found most helpful - a book titled "SQL Server 2000 Web Application Developer's Guide". It's biased toward ASP and Microsoft's Visual InterDev - which were standard development tools here at the time. I haven't searched yet for a comparable book for .NET, but would bet one exists.
As for your "best" option? I tend to base that on the tools I have available and the time constraints I'm under. There are many different tools and languages that will get you there - it just depends on the resources you have to put towards it and the life expectancy of the system.

It is actually very easy leave the Access frontend alone and create the Web frontend, if you are in .NET 1.1 it is more difficult but if you have not bought tools then you can just download the free Visual Web developer Express and use the free training Videos provided by Microsoft. You also have the option to use existing Starter Kits that are almost complete applications you can just add what you need. Try the links below for the free tutorials, Videos and Starter Kits, the second link is more free training from the Asp.net founder's blog. Get started and you will see it is not complicated. BTW you can also post questions at the forums, post again if you still have questions will be glad to answer them for you. Hope this helps.
