You need to create a share on the folder above, you should not use the administrative share. Use either a local user account or a domain account...

We have Share Point 2010 databases on SQL Server 2008R2. The full backup of all content databases is taking 12 hours (total data size 1.5 TB)

Have you tried extracting the SP to a folder on the C drive and expecting the setup from there?

3) Full database backup must be taken if the above statements are execute as they break the log chain.
A differential backup is sufficient to restart the log chain...

Is having mix of drive letters and Mount points in a single cluster supported?
It makes no difference either way. The currently assigned disks with their letters will be dedicated...

SSRS is not cluster aware under MSCS, although you could look at Veritas Cluster Server to achieve high availability here.
