In term of capacity, There is no problem. But you shouldn't use developer edition for your production system.

Kill the user's connection is not the solution to resolve the backup failure. We are running 7X24 and get backup done well.

Full backup will backup everything for you. It is good practice to script all SPs too.

File backup just backs up the physical database files (MDF).
