You can use Cross tabs.
SELECT Region, District, LocID,
MAX(CASE WHEN Title = 'IT Director' THEN Cnt ELSE 0 END),
MAX(CASE WHEN Title = 'IT Manager' THEN Cnt ELSE 0 END)
--etc
FROM #Loc
GROUP BY Region, District, LocID;
