If I have two instances running on a server and the instances are both named instances.
Do I have to do some special configuration of SQL browser, or leave as default should be OK?

You shouldn't have to do anything, it already knows about both instances.
Where you might have issues is if you have a firewall up on the SQL machine it is possible that could block access to the browser.
