I have one System (box)on which I have two databases which are to be mirrored on another instance on the same system(computer/box) i.e DBprincipal1...............DBMIrror1
DBPrincipal2...............DBMirror2
Now can I use same endpoint for configuring two mirror sessions or will I require another mirror endpoint?
Another scenario where in if second instance is on the another box which will have mirror databases then how endpoints to be configured?and what about port number setting can I use same port number or has to be different for both the partners?

you need an endpoint on each instance in the mirror relationship, if they're on separate machines you can use the same port number.

Will two mirroring sessions on same instance use single endpoint or I require
two separate endpoint for two mirroring session on my box.

IIRC an instance can have only one mirror endpoint and that multiple sessions will share that endpoint.
You will need to check Books Online to be sure as it has been several years now since I dealt with mirroring.

The endpoint is instance wide so both will use it
